Antipyretic and analgesic action proposed to be via central inhibition of prostaglandin synthesis (COX-3 variant in CNS) and modulation of serotonergic, cannabinoid, and TRPV1 pathways. Lacks meaningful peripheral COX inhibition, hence minimal anti-inflammatory effect and reduced GI toxicity relative to NSAIDs.
Mild to moderate pain in dogs (often combined with codeine 1-2 mg/kg PO). Useful when NSAIDs are contraindicated (GI ulceration, renal insufficiency). ABSOLUTELY CONTRAINDICATED in cats and ferrets (deficient UGT1A6 glucuronidation → NAPQI accumulation → methemoglobinemia and hepatic necrosis; lethal at ≥10 mg/kg).
