Leucovorin (calcium folinate) is a reduced form of folic acid that does not require dihydrofolate reductase for activation. It bypasses the metabolic block caused by methotrexate and other folate antagonists, rescuing normal cells from antifolate toxicity while maintaining anticancer efficacy.
