Afoxolaner is an isoxazoline ectoparasiticide that acts as a non-competitive GABA (gamma-aminobutyric acid) receptor antagonist. It binds to chloride channels in insect/tick nerve and muscle cells, blocking neuronal signal transmission. Affected parasites are paralyzed and die. Significantly more selective for insect GABA receptors than mammalian receptors, providing a wide safety margin.
Treatment and prevention of flea (Ctenocephalides felis) and tick (Dermacentor variabilis, Ixodes scapularis, Amblyomma americanum, Rhipicephalus sanguineus) infestations in dogs. Also used off-label for sarcoptic/demodectic mange.
