Anticancer agent. Doxorubicin is derived from the soil fungus Streptomyces peucetius. Doxorubicin damages DNA by inhibition of the enzyme topoisomerase II. This enzyme is responsible for DNA functions. When topoisomerase is inhibited, the DNA segments cannot perform transcription, leading to breaks in the DNA strands and cell death. Secondarily, this mechanism causes cell death by blocking synthesis of RNA and proteins. Doxorubicin also forms free radicals (OH) that can attack DNA and lead to oxidation of DNA. Doxorubicin is used for treatment of various neoplasia, including hemangiosarcoma and lymphoma. Doxorubicin is commonly used in humans for the treatment of breast tumors, various sarcomas, and osteosarcoma. In veterinary medicine it has been used for lymphoma, osteosarcoma, and other carcinomas and sarcomas. It is considered one of the most effective single agents in the treatment of lymphoma. It is used commonly in cancer protocols with other agents.
